我需要从一开始对设计好的primer进行检测开始的整个实验过程的protocol,
用的是cyber green.

ABI的机器，主要是我不知道一开始应该怎么条件设定，比如template的浓度阿什么的，说明书上写的只有多少多少微升，浓度阿什么的没有写。
另外就是手头上的primer要具体作哪些预备试验来确定是可以用的primer。

设计了引物融解曲线，看产物是否单一（不要有杂带产生）。机器的话应该是有手册的，问负责机器的人要一下看有没有。

你是用那个公司的机器？
BioRad 有本小册子，写得很详细，你可以问他们要一下。

Primers can be designed by ABI's program. The principle is there is not other PCR band. CYBER green is just DNA intercalation chemical, it binds any double strand DNA. Therefore, specific primers are critical. Otherwise, fluorescence labeled (such as FAM-labeled probe) probe should be used.
