本实验通过对三角帆蚌取血进行血液涂片观察,根据血细胞的形态结构特点,将三角帆蚌的血细胞分为透明细胞、颗粒细胞和成纤维细胞,其中颗粒细胞根据其大小又分为小颗粒细胞、大颗粒细胞,根据其染色性又分为嗜酸性颗粒细胞和嗜碱性颗粒细胞.以M199培养基和RPMI1640培养基为基础,利用不同的培养条件,对血细胞进行原代培养,结果表明,以M199培养基为基础,添加20%的胎牛血清、5%的1×Hank`s平衡盐、100ug/mL双抗和支原净,培养温度28～30℃,pH值7.7.2的条件下培养效果较好,培养24h后细胞分裂速度加快,20%的细胞贴壁生长,贴壁细胞形状各异,生长可维持7天左右.悬浮细胞占80%,细胞活力强,培养14天后,仍能传到第二代.
